OpenSPIM is an Open Source method of Light Sheet microscopy using off the shelf and customized components. Nobska Imaging can help you build your own system or upgrade your existing system.
The SPIM innovation offers quick optical sectioning, removing out of focus data. It accomplishes this by centering a 3-5um light-sheet through the sample, taking two-dimensional pictures of the illuminated plane with a detection objective and camera at 90 degrees to the illumination plane. Three-dimensional stacks are assembled by moving the sample through the static light-sheet. The sample is mounted in low melt agarose or hydrogel, and drawn into a syringe. The syringe is mounted in an XYZ rotation stage and placed carefully into the chamber. The agarose is then pushed into the beam and held by the syringe or a custom 3D printed holder.
